5th for me... ( depending on the dyno the 4th can show too much hp. )

You want it to be as close to 1:1 as possible, so why on earth are you doing it in 5th. God that must take forever...not to mention the strain on the bottom end.

in practice Ive seen differences up to 20hp. ( between 4th an 5th. )

thats what pretty much everyone around here ( germany ) does.
depending on the dyno: on 4th gear the run is way too fast -> messes up the result ( as mentioned above )

*edit*
the gearing on my 03 cooperS is:
| 1 | 11.425
| 2 | 7.181
| 3 | 5.397
| 4 | 4.407
| 5 | 3.656
| 6 | 2.986
| R | 11.130

so if you want to do the dyno in the gear as close to 1:1 as possible, you should be using the 6th.
