Must drive roads
The Going to the Sun Road in Glacier National Park.
It is suppose to be one of the greatest roads of all time, not a super high speed run. But, a nice VERY scenic drive through some very beautiful country. I have not personally done it but I would love to.
Also if you are in the NorCal area I very much advise motoring on Caleveras Road from Sunol to Milpitas.

A little more CenCal than NorCal is the 58 and the 229.
But the Santa Cruz Mountains are my favorite thats where I learned how to drive, where had my first accident/s (plural) Here they are look at the 35 (summit/skyline to locals), the 84, and the 9, also my favorite the 236 (where I totaled my first car a 76 Monte Carlo splat against the side of the hill) Pan left for the 236 and the 84 and in for a closer look of these roads.

I drove CA-35 (Skyline Blvd) from end-to-end today. Nice, windy road without being neck-breakingly (or carsickeningly) twisty. Blasphemy, I know 

It was raining on the south side of the range, but sunny on the north side. There are a few "scenic overlook" stops on the way. I stopped at the first decent-sized one to admire the view and have a little picnic

I have found that Hwy 1 is not to be missed.
Also: Hwy 49 end to end
Hwy 299 and Cal 36 and a personal favorite; Road 600 off of 49 near oakhurst
Dont miss hwy 25 from Hollister to Hwy 198 and also Carmel Valley Road (G-16)

Done 'em all except Road 600. Another good one is Hwy 155 east out of Delano.
Kancamagus Highway (Rt. 112)/Bear Notch Road, in New Hampshire.

Death Valley California.
Starting in Pahrump Nevada head to Armogosa then to Furnace Creek. Lots of ways to go from there and explore.........into Beatty, left onto the Valley floor..... etc. Stay at the Inn in Furnace Creek and take a week to explore.

Does Hawaii count? The road past the "mini Grand Canyon" on Kau'ai up to the lookout over the Na Pali coastline is great fun and quite a good length. I was fortunate to have a good handling hire car (now there's a rarity!

If you're ever in Middle Tennessee here's a few to try out.
US 231 between Shelbyville and Fayetteville is a good mix of curves, straights and changes of altitude.
Another good twisty with some straights mixed in is SR 30 between SR 70S and I-75 makes for an interesting drive. While you're on this route take time to go to Fall Creek Falls State Park and Forest. Very scenic with the second tallest waterfall east of the Mississippi.

I tried one this summer and would not recommend one for a spirited drive. In many places it is just too narrow for two cars. So you have to be slow and careful. If it does not stop you then - half of the road was covered by gravel, so you are accompanied by the sound of them hitting your car's body. Hate this.
I saw a closed turn ahead of me and slowed down enough as I didn't know what's behind it (fast oncoming car is pretty much enough to total both cars as I mentioned it was too narrow in many places). No it was not the car. It was a bike. Front wheel off th ground. I just had enough time to estimate on where particularly on the hood (I wasn't in a Mini) his front wheel will land. He didn't have steering so he was going straight and so it was easy to know where he will hit me. The guy was completely realizing how badly wrong he was. Luckily for me the bike was not powerful and heavy enough. Sort of the one for off road. So he tried his best to brake and not to get into a complete slide, land his front wheel, brake, turn, brake... Whew... We are safe. I could only help him by bringing the car to a complete stop to give him more space, and stay there scared by the possibility to be rear ended by another car. Luckily that didn't happen. I saw other fun to drive cars there, but they all maintained reasonable speed.
