I don't know the ratios, but i do believe that it would slow you down a bit...unless you have some mods. If a 6spd getrag could be fitted instead of a 5spd midlands, then I would love to know how! If that would work, then I'm sure a 5spd getrag would work. I would prefer a 6spd also, but I just think that 4-6 would be a bit to big for a stock cooper engine. I'm sure 1st is very similar since there is little or no boost at 2k rpm. You have to be able to comfortably get going from a stop. My concern would be using the bigger gears for passing. Were talking about a 50 hp difference.

Again, I don't have the ratios in front of me. If anyone has any information on HOW to make the midlands to getrag swap work...I would love to know.

You can ask Richard22b who owns the Mini One with the 6spd conversion any questions here.

Or I'll try and get him to come on here

From what I understand, the Getrag 5 and 6 speeds have the same final drive ratio, so if the 5 speed works on a Cooper, a 6 speed should work, as well. More ratios in the same range should be a good thing.

I think the six speed would be more of a plus for the Cooper than it is for the Cooper S.

I'm not sure how accurate this information is, but mini2 has an faq article with gear ratios in it.

According to those numbers, they are taller in the 6spd, although not by much, mainly in 1st and 2nd. I'm curious about how this drives. There isn't that much difference after 1st. At least I have a couple options other than shipping an uprated tranny from overseas now.
